研究概述
在《物種起源》中,達爾文提出"適者生存"的重要概念。"適者生存"的關鍵是成功應對生存威脅。微觀尺度上的生存威脅主要是病原微生物。它們可以入侵人和動物的消化道和呼吸道,引發一系列病理性的防御反應(如惡心和嘔吐)。宏觀尺度上的生存威脅主要是天敵。在面臨天敵威脅時,人和動物表現出一系列生理性防御反應(如戰斗和逃跑)。大腦如何感知這些不同尺度的生存威脅并啟動相應的防御反應,是神經科學領域一個重要的科學問題。我們實驗室以小鼠為動物模型,在微觀尺度上,研究了大腦啟動惡心和嘔吐的神經機制;在宏觀尺度上,研究了大腦啟動戰斗和逃跑的神經機制。
研究內容-1: 大腦感知病原入侵啟動防御反應的神經機制
食物中毒引發的惡心和嘔吐是機體自我保護的防御反應,對生存具有重要的意義。惡心和嘔吐也是癌癥化療的主要副作用,具有重要的臨床意義。在過去三十年里,惡心和嘔吐的機制研究一直舉步維艱,因為領域內缺乏合適的動物模型和研究范式。
2022年11月1日,我們在Cell上發表了題為The gut-to-brain axis for toxin-induced defensive responses的研究論文。該研究建立了以小鼠為動物模型研究惡心和嘔吐反應的新范式,發現腸道報告病原入侵的“情報員”細胞,迷走神經傳輸情報的“線人”細胞,以及腦內負責啟動惡心和嘔吐反應的“開關”細胞。在分子細胞層面上,揭示免疫反應和5-羥色胺信號在引發惡心和嘔吐中的重要功能,證明化療藥物是通過“綁架”這些分子細胞機制引發惡心和嘔吐的副作用。研究還發現,這些分子細胞和神經環路機制也參與了化療藥物(doxorubicin)引發的“惡心-嘔吐”副作用。這一發現暗示,化療藥物引發的“惡心-嘔吐”副作用,可能是通過綁架進化上保守的“食物中毒”機制得以實現。
同期的 Cell 配發了專家評論文章,對這一工作進行了詳細介紹。研究成果還被Nature Reviews Neuroscience以Research Highlight的形式進行了報道。紐約時報也對這一研究成果進行了采訪和報道。
研究內容-2:大腦感知天敵威脅啟動“戰斗-逃跑”反應的神經機制
人和動物在面臨生存威脅(例如天敵)時,可以表現出“戰斗-逃跑”行為反應,《水滸傳》中的武松打虎便是這一過程的生動范例。在某些人群中,過度或反復的“戰斗-逃跑”行為反應引發一類稱為創傷后壓力應激障礙(PTSD)的精神疾病。要揭示一類疾病的發病機制,往往需要從源頭入手分析。解析引發“戰斗”和“逃跑”行為反應的關鍵神經環路可能是深入分析PTSD發病機理的突破口。近年來,我們在這一研究方向上取得一系列研究成果。
我們在實驗室里建立了小鼠與天敵“戰斗”的行為范式,并發現天敵攻擊時產生的傷害性機械刺激是觸發小鼠戰斗行為的關鍵。我們在小鼠的下丘腦前核 AHN中發現編碼傷害性機械刺激的神經元。光激活這些神經元,能直接引發小鼠對抗天敵的防御行為。抑制這些神經元,小鼠失去了對抗天敵的能力。這一研究成果于2022年1月3日發表在Nature Neuroscience上。
大腦如何啟動“逃跑”的行為反應呢? 我們在小鼠的大腦中發現一群啟動逃避天敵行為的神經元。這些神經元位于一個叫做“上丘”的腦區,以小清蛋白(parvalbumin,PV)為生物標記物。上丘PV神經元可以像雷達一樣監測天空中快速逼近的運動目標,并把這些預警信息傳送給恐懼中樞杏仁核。直接刺激該神經環路,可以快速引發強烈的逃跑行為。反復刺激該神經環路,動物出現類似PTSD的核心癥狀。這些科學發現于2015年6月26日發表在Science上。
以上關于“戰斗”和“逃跑”行為反應的神經機制的研究結果為進一步深入研究PTSD的發病機理奠定了重要基礎。
